World hazelnut production is based on European hazelnut (Corylus avellana) and is limited by the narrow climatic requirements of this species. The cold hardiness and disease resistance of the American hazelnut (Corylus americana) offer opportunities to expand production to new areas including the Upper Midwest (USA). The American hazelnut is a phenotypically diverse species. This study used ten microsatellite marker loci to investigate genetic diversity in 1140 individuals sampled from 25 populations across Wisconsin, Iowa, Minnesota and North Dakota. Overall, the marker loci were highly polymorphic (Ho = 0.69, He = 0.78, PIC = 0.84) with 7–13 alleles per locus. There was very high genetic diversity within populations (90% of the total) and some tendency toward population differentiation. Mantel’s test showed that genetic distance among the populations was not correlated with geographic distance. We conclude that selection of individuals for use in breeding should be based primarily on phenotype (productivity, nut size, percent kernel, ease of harvest), with care to include representatives of genetically differentiated populations. 相似文献
Medical records of 23 dogs with histologically documented orbital neoplasia and admitted to the New York State College of Veterinary Medicine between 1975 and 1984 were reviewed. Almost all (91%) of the tumors were classified as malignant; 74% of the tumors arose as primary neoplasms within the orbit. Eleven tumor types of connective tissue, bone, epithelial, and hemolymphatic origin were represented. The typically afflicted dog was purebred, female, and middle-aged. Review of this series confirmed the clinical impression that orbital neoplasms in dogs are aggressive malignancies with poor long-term prognosis. 相似文献
OBJECTIVE: To compare trochlear block recession (TBR) to trochlear wedge recession (TWR) with regards to patellar depth (percentage of patellar volume under the trochlear ridges), patellar articular contact, percentage of recessed trochlear surface area, and resistance to patellar luxation. STUDY DESIGN: In vitro computed tomography (CT) and biomechanical evaluation using a cadaver model. SAMPLE POPULATION: Twelve normal, large-breed canine cadavers. METHODS: Bilateral pelvic limb specimens with intact stifle joints were mounted on a positioning device. The femoral trochlear ridges were reduced to provide a standard shallow trochlea. TBR or TWR was performed to a standard depth randomly on paired specimens. CT and biomechanical evaluations were performed pre- and postoperatively in both an extended (148 degrees ) and flexed (113 degrees ) stifle position. CT images were digitized and measurements made using an image-analysis software program. Biomechanical testing consisted of applying 40 degrees of internal tibial rotation and documenting patellar luxation. RESULTS: The change in trochlear depth (depth of recession) was not significantly different between groups. In the extended stifle position (patella in the proximal trochlea), patellar depth and patellar articular contact with the recessed trochlea were significantly greater after TBR compared with TWR. The percentage of recessed trochlear surface area was significantly greater after TBR compared with TWR. In the extended position, a smaller percentage of the patellae luxated within 40 degrees of internal tibial rotation after TBR compared with TWR. CONCLUSIONS: TBR increases proximal patellar depth, increases patellar articular contact with the recessed proximal trochlea, recesses a larger percentage of trochlear surface area, and results in a greater resistance to patellar luxation in an extended position as compared with TWR. The spread of tomato yellow leaf curl virus (TYLCV) is significantly correlated with the population size of its vector,Bemisia tabaci Genn. The perennial weedCynanchum acutum L. and the annual weedMalva parviflora L. were found to be natural hosts of TYLCV in the Jordan Valley.C. acutum is not a preferred host forB. tabaci, but the whitefly feeds on it sufficiently long to acquire the virus. Whiteflies marked with fluorescent dust while feeding naturally onC. acutum along the banks of the Jordan River, were subsequently trapped within the main tomato-production area 7 km away. An increase in theB. tabaci population and in TYLCV infectivity was found in plots surrounded by windbreaks. We established Populus deltoides Bartr. stands differing in nitrogen (N) availability and tested if: (1) N-induced carbon (C) allocation could be explained by developmental allocation controls; and (2) N uptake per unit root mass, i.e., specific N-uptake rate, increased with N availability. Closely spaced (1 x 1 m) stands were treated with 50, 100 and 200 kg N ha(-1) year(-1) of time-release balanced fertilizer (50N, 100N and 200N) and compared with unfertilized controls (0N). Measurements were made during two complete growing seasons from May 1998 through October 1999. Repeated nondestructive measurements were carried out to determine stem height and diameter, leaf area and fine-root dynamics. In October of both years, above- and belowground biomass was harvested, including soil cores for fine-root biomass. Leaves were harvested in July 1999. Harvested tissues were analyzed for C and N content. Nondestructive stem diameter and and fine-root dynamic measurements were combined with destructive harvest data to estimate whole-tree biomass and N content at the end of the year, and to estimate specific N-uptake rates during the 1999 growing season. Shoot growth response was greater in fertilized trees than in control trees; however, the 100N and 200N treatments did not enhance growth more than the 50N treatment. Root biomass proportions decreased over time and with increasing fertilizer treatment. Fertilizer-induced changes in allocation were explained by accelerated development. Specific N-uptake rates increased during the growing season and were higher for fertilized trees than for control trees. 相似文献
OBJECTIVE: To determine if preoperative topical administration of ciprofloxacin or ofloxacin results in aqueous humor drug concentrations that exceed the MIC(90) of common ocular contaminants in dogs undergoing cataract surgery. PROCEDURES: Twelve dogs were treated with topical 0.3% ciprofloxacin and 13 dogs with topical 0.3% ofloxacin once the night before surgery, and then with 1 drop of ciprofloxacin or ofloxacin every 15 min for 2 h immediately before surgery. Aqueous humor samples were taken from each eye immediately before the incision was made and frozen at -70 degrees C. First eye samples (S1) were taken closer to the time of topical treatments than second eye samples (S2). Samples were analyzed by high performance liquid chromatography (HPLC) at the North Carolina State University (NCSU) Clinical Pharmacology laboratory. RESULTS: In ciprofloxacin patients, S1 concentrations were 0.03-0.69 (0.17 median) microg/mL, and S2 concentrations were 0.09-0.95 (0.36 median) microg/mL. Aqueous humor concentrations did not exceed the MIC90 of Streptococcus sp. Few eyes (1/12 OU) exceeded the MIC90 for Staphylococcus sp. or Corynebacterium sp.; moderate numbers (5/12 S1; 8/12 S2) exceeded the MIC90 for E. coli. In ofloxacin patients, S1 concentrations were 0.48-2.81 (1.05 median) microg/mL, and S2 concentrations were 0.45-3.63 (1.30 median) microg/mL. Although few eyes (相似文献
The aim of this study was to determine the effects of various abiotic factors, such as light, physical stress (pipetting) and thermal shock, on the quality of fresh and cooled equine sperm. In experiment I, four sperm aliquots were subjected to different light exposures: (i) protected control samples (CTRL), (ii) exposed to UV light at 10 cm (UV10), (iii) exposed to UV light at 20 cm (UV20) and (iv) exposed to laboratory lighting (LAB). In experiment II, four semen aliquots were subjected to repeated pipetting for 0, 10, 20 and 30 times (CTRL, P10, P20 and P30, respectively). In experiment III, four semen aliquots at 15°C were subjected to thermal oscillations: (i) cooled control sperm at 15°C (CTRL), (ii) oscillations of 1.9°C/min to a temperature of 30°C (T30), (iii) oscillations of 1.4°C/min, with the temperature rapidly falling until reaching 1.3°C (T0R) and (iv) oscillations of 1.1°C/min, with the temperature slowly falling until reaching 4.2°C (T0S). The results revealed that after 30 min, UV10 and UV20 sperm samples showed significantly (p < .05) lower total and progressive motility values, sperm kinematic parameters and mitochondrial potential. After 45 min of exposure, differences were highly significant (p < .001). No significant differences (p > .05) were found for pipetting or thermal oscillations. The results suggest that, even if equine sperm samples are not handled in the laboratory under optimal conditions, fresh and cooled equine spermatozoa are able to resist the impact of various abiotic stimuli without any reduction in their quality. This study analyses the effect on normospermic samples, but future research could look at the tolerance that asthenozoospermic equine samples have to these abiotic influences. 相似文献
AIMS: To assess, in dairy cows between 60–160 days of gestation, whether the agreement between actual gestational age and that predicted by placentome size could be improved by using individual placentome measurements rather than a single average, and to identify how increasing the number of placentomes measured improved the prediction.
METHODS: Data were obtained from 58 cows examined using transrectal ultrasonography every 10 days between 60–130 days, and every 15 days between 130–160 days of gestation. For each cow, four to six placentomes in each of the pregnant and non-pregnant horns were examined from the region of the uterus near to the cervix. A mixed-effects model, which included cow as a random effect, and a simple linear regression which ignored within-cow correlation, were fitted to the data. Inverse regression was used to compare the 95% prediction bands obtained for estimating gestational age using the means of three, five or eight placentome measurements.
RESULTS: The fit of the mixed effects model was better than a simple linear regression (p<0.001) but the fitted lines from the two models were very similar. Using the simple regression model, for a mean placentome length of 15?mm estimated gestational age was 100 days, with 95% CI of 68–131 days for measurement of three placentomes, and 80–120 days for eight placentomes.
CONCLUSIONS: The agreement between placentome size and gestational age was improved by increasing the number of placentomes measured. Direct comparison of these CI with those for other published fetal measures was not possible as similar prediction bands have not previously been calculated for fetal measures; however one study reported a residual SD which had been calculated using size measurements as the predictor variable and age as the response variable. Using these data 95% CI were calculated to be ±9 days for crown-rump length and ±25 days for uterine diameter. These are likely to be an underestimate of the true CI and do not take account of the increase in variance of the difference between predicted and actual gestational age as gestational age increases.
CLINICAL RELEVANCE: Placentome measurement may be useful for estimating fetal age in late gestation. Further research is required to better establish the agreement between gestational age and the fetal measurements which are currently used to estimate fetal age. 相似文献
